Abstract (EN)

The presence of autoantibodies against intracellular antigens is an important indicator of systemic connective tissue diseases. In the Indirect Fluorescent Antibody (IFA) method, nucleus and cytoplasmic autoantibody positivity show different patterns of staining that can be clinically associated with diseases. In this study, it was aimed to investigate anti-DFS-70 antibodies by Immunoblot method in serum of patients with Anti-Nuclear Antibody (ANA) positivity in granular + granular chromosome pattern by IFA method. Antibodies against 16 different antigens were investigated by immunoblot method in 100 serum of patients from IFA and granular + granular chromosome patterns of patient sera sent from various clinics of Gaziantep University Medical Faculty Sahinbey Research and Application Hospital between April 2016 and September 2016. Demographic information of the patients included in the study was recorded. Between April 2016- September 2016, 4440 serum samples from various clinics were evaluated in terms of ANA by IFA method and alone granular + granular chromosome style ANA positivity in 81 samples (1.82%), combination with other pattern Granular + granular chromosome-like ANA positivity in 19 samples (0.42%) has been determined. 100 samples determined positivity studied by immunblot method and anti-DFS70 antibody positivity as single in 70 (70%), different antibody positivity with anti-DFS70 positivity in 30 (30%) has been detected. Anti-centromere positivity has not been detected in any patients. ANA positivity in the granular + granular chromosome pattern by the IFA method is associated with the presence of anti-DFS70 antibody that our results confirm this. However, in cases where there is an ANA positivity against more than one antigen, it has seen that IFA staining patterns could mask each other. It was concluded that immunoblot method is useful in the presence of more than one antibody.
