An in vitro cytotoxicity of indirect composite materials before and after aging them in artificial salivaThe purpose of the current study was to investigate the effect of storing indirect composite materials in artificial saliva.Disk shaped specimens (10 mm diameter, 1mm thick) were polymerized using the dentacolor XS light source (Kulzer, Wehrheim, Germany). 50 specimens were fabricated for each indirect composite material. Specimens (Artglass n=25, Solidex n=25) were divided into two groups: one for storing in artificial saliva and the other for direct testing. The first group (Artglass n=25, Solidex n=25) was stored in artificial saliva at 37 ºC for 48 h. Being removed from the artificial saliva, the specimens were aged for 1h, 24h, 72h, 1 and 2 weeks in Dulbecco?s Modified Eagle Medium/Ham?s F12 (DMEM/F12). The second group (Artglass n=25, Solidex n=25) was aged for 1h, 24 h, 72 h, 1, 2 weeks in DMEM/F12. After both groups were aged, cytotoxicity of their extracts in cultured fibroblasts (L929) was measured by MTT assay. The degree of cytotoxicity for each specimen was determined according to the reference value represented by the cells with a pure culture medium. Student?s t Test was used to the determine whether or not distribution of living cell percentage measurements were normal.Generally, the tested materials stored in artificial saliva for 48 h demonstrated higher cell survival rates when compared with the non-stored tested materials that had not been kept in artificial saliva for all experimental times.This findings suggest that composite resins used in indirect dental restorations differ in their cytotoxic potential and their ability to affect cell survival rates.Key words: cytotoxicity, indirect composite resin, MTT
