Toxic effects of indoxacarb on MDA-MB-231 breast cancer cell line

Abstract (EN)
Indoxacarb is an insecticide from the oxadiazine group and is widely used for the control of insects in agricultural areas. Breast cancer is also the first type of cancer seen in women in our country. In this study, the effects of increasing doses of indoxacarb applied to breast cancer cell line MDA-MB-231 cells were investigated using the MTT method (3-(4,5-dimethyltriazol-2-yl)-2,5-diphenyltetrazolium bromide) and the comet method. (MDA) level and antioxidant enzyme activities were also investigated. The cytotoxic effect of indoxacarb on MDA-MB-231 cells was found to be 45 µM in the control group and administration groups (10-120 µM). According to the results of the cytotoxicity analysis, it was determined that the cell viability decreased as the indoxacarb concentration increased. For the detection of DNA damage in cells, DNA damage was determined depending on the dose increase in DNA tail percentage, tail length and tail moment values in the control group, 22,5 µM, 45 µM, 90 µM indoxacarb applied groups according to the comet analysis. Malondialdehyde (MDA) level and antioxidant defense system enzymes; Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities were evaluated comparatively by measuring control and application groups in spectrophotometer. While an increase was observed in the amount of MDA, a decrease was observed in antioxidant enzyme activities. In the light of these studies, the doses that indoxacarb caused toxic effects on the human breast cancer cell line MDA-MB-231 in vitro were determined within the scope of the studied parameters. The present study is a preliminary study on this subject and will contribute to the literature and will shed light on future comprehensive studies.
