Abstract (EN)
In this study, it is aimed to define protective and restorative effects of Momordica charantia (bitter melon) extracts (MCE), which are commonly used in pythotherapy aganist DNA damage inducted with hydrogenperoxide, in Saccharomyces cerevisiae, from eucaryotic cells, in vivo contitions. In this purpose, haploid Saccharomyces cerevisiae BY4741 (MATa his 3Δ1 leu2 Δ0 met15Δ0 ura3Δ0) strains were used. While protective and restorative induction effects of the active substance utilized were defined by using alkaline single cell gel electrophosis (comet assay) method, antioxidative effects were analyzed by measuring total antioxidant status (TAS) and total oxidant status (TOS). MCE was appliedto S. cerevisiaecells in of 1/16, 1/32 and 1/64 dilusions. Our results showed that MCE protectsth esecells aganist DNA damage formed with hydrogenperoxide (H2O2) in S. cerevisiae. It is found as a result of the application that the active substance has a protective effect on the cell in 1/16 dilusion. MCE in dilusions of 1/16, 1/32 and 1/64 was found to be insufficient in restoring DNA damage.
