The evaluation of the favourable effect of Bevacizumab on classical BEP treatment in Human Testicular Germ Cell Tumour: An In vitro study

Abstract (EN)

Testicular cancer constitutes 13-23% of tumors of the male urogenital system. Testicular tumors are the most common malignant tumors in men between the ages of 20 and 40 and 90-95% of them are germ cells. A combination of BEP (Bleomycin, Etoposide, Cisplatin) is commonly used in the treatment of human testicular germ cell tumor. Although this combination has a therapeutic efficacy of 90%, the high incidence of side effects has necessitated the development of new treatment protocols. Bevacizumab is an anti-tumor drug that is used in the treatment of many tumor types. The aim of our study; The aim of this study is to evaluate the potential or inhibitory effects of anti-VEGF bevacizumab which is an increasingly common chemotherapeutic agent that can potentiate BEP therapy. In our study, after determining the IC50 dose in the ACC 811 Human Germ Cell Tumor cell line in combination with BEP in the cell culture medium, this dose was calculated by using MTT in these combinations by combining these doses with different bevazicumab doses. Immunocytochemistry staining was performed to determine the VEGF, Caspase-3, Bax and Bcl-2 expressions in the samples. After that, immunopositive and immunonegative cells were counted and the results were statistically analyzed. As a result; Although bevazicumab has a cell-killing effect in the GCTT cell line, we believe that this effect is effective through a further pathway activation or inhibition that is not performed by the Bax, Bcl2 and Caspase 3 pathways. Decreased anti-VEGF effect on VEGF at higher doses has shown us that bevazicumab cannot be very effective in this tumor type but it should be supported with advanced and broad spectrum studies.
