Research on probiotics characterization some of Lactobacillus species bacteria isolated from human vagina

Abstract (EN)
In this study, 58 strains of Lactobacillus species bacteria were isolated from 10 patients out of 19 women who were known to be healthy and/or in pregnant, HPV(+), in menopause, having an infection, in post delivery period or having irregular menstruation. As a result of the identification tests the bacteria isolated were identified as 12 L. gassed, 9 L. acidofilus, 9 L. vaginalis, 8 L. er ispat us, 8 L. delbrueckii spp. lactis, 3 L. plantar urn, 2 L.jensenii, 2 L. salivarus, 2 L. cellobiosus, 1 L. brevis, 1 L. curvatus and 1 L. oris. Amount of acid and hydrogen peroxide production of Lactobacillus species was estimated. The amount of acid produced was found to be between 0.27-1.33 %. The amount of hydrogen peroxide produced was found to be between 1.01-15.50 ug/ml with the spectrophotometric method, where as qulitatively 41% of it was estimated to be double positive, 19% of single positive and 40% of it was negative in agar with tetramethylbenzidine. The antimicrobial effect of Lactobacillus was studied by using the agar diffusion technique. Lactobacillus had an inhibitory effect on the 85% of P. aeruginosa ATTC 29212 strains, on the 48% of S. aureus ATTC 2392 strains, on the 71% of S. enteritidis RSKK 171 strains and on the 43% of E. coli ATTC 11230 strains. An antimicrobial effect on the C. albicans AJD 180 ve C. albicans ATCC 10239 strains could not be observed. None of the Lactobacilli were found to have a bacteriocin effect on the test bacteria.IV susceptibility tests of isolated Lactobacillus species to amoxycillin (25 ^g), azithromycin (15 p.g), chloramphenicol (30 \ig), teicoplanin (30 fig), ciprofloxacin (5 fig), clarithromycin (15 fig), streptomycin (10 |xg) and tetracycline (30 fig) were determined using the disk diffusion method following NCCLS standards. In the results of the research, most of the Lactobacillus species were found to be highly resistant to ciprofloxacin (98%) and streptomycin (93%) and most sensitive to clarithromycin (95%) and tetracycline (88%). In addition, the inhibitory concentrations of isoconazole and oxyconazole that were used as antifungal drugs were determined on C. albicans AJD 180. The inhibitory effect of both antifungal drugs at 0.2; 0.1 and 0.05 |ig/ml concentrations on Lactobacillus isolates were investigated by using the agar diffusion method. At the end of the study, although both antifungal drugs showed a highly inhibitory effect on C. albicans AJD 180, it was determined that oxyconazole had a very low inhibitory effect on Lactobacillus isolates.
