Histopathological examination of TRPM8 ion channel expression in intracranial glial tumors and correlation with adc measurements in diffusion MRI

Abstract (EN)
Transient Receptor Potential Melastatin (TRPM) ion channels are cation channels in the plasma membrane. The expression and effects of TRPM ion channels in glial masses are not clearly known. Magnetic Resonance Imaging (MRI) is the imaging method of choice for the diagnosis of glial masses. In our study, TRPM8 levels were measured in glial masses, and the Apperent Diffusion Coefficient (ADC) signal differences in tumor, tumor periphery and symmetrical brain tissue were evaluated in Brain Diffusion MRI. Two groups were formed with 30 patients diagnosed with an intracranial glial mass that we underwent surgery. In Group A; 15 with low-grade glial masses were in Group B; There were 15 patients with high-grade glial masses. The control group, Group C, was formed from brain tissues taken from 10 patients who had previously been operated for other reasons such as cerebral hemorrhage. TRPM8 was examined immunohistochemically in tissues. In Diffusion MRIs of group A and B patients, ADC measurements were made within the mass, around the mass, and symmetrical brain tissue. The obtained data were analyzed in SPSS (Statistical Package for Social Sciences) program. Significant increase in TRPM8 immunoreactivity was observed in Groups A and B compared to Group C. A significant increase in TRPM8 was observed in Group B compared to Group A (p=0.005). In Group B, ADC values were lower in measurements on the mass compared to Group A. Mass circumference values were higher in Group B compared to Group A. There was no significant difference in symmetrical brain tissue measurements. In high-grade glial masses, that is, more malignant and hypercellular glial masses, TRPM8 expression and activation transport were more pronounced. As the tumor stage increased, the decrease in the mass ADC values, that is, the decrease in diffusion, was associated with hypercellularity. As the tumor grade increased around the mass, the increase in ADC values was associated with an increase in peritumoral edema.
