Effect of intracerebroventricular nesfatin-1 injection on ghrelin and leptin levels in male rat

Abstract (EN)

Nesfatin-1, a newly discovered neuropeptide, is an anorexigenic peptide produced in the hypothalamic areas. Stress response and hypothalamo-pituitary-adrenal (HPA) axis activation also play an important role in the anorexigenic effect of this peptide. In the regulation of the stress response, in addition to corticotropin-releasing hormone (CRH1 and CRH2) receptors, the oxytocin receptor also plays a role in nesfatin-1 depending effects. Leptin is an anorexigenic peptide produced from adipose tissue and join to the circulation. Ghrelin is an orexigenic neuropeptide produced by the gastrointestinal tract. Asprosin is a recently discovered peptide that is released during fasting and increases hepatic glucose production. Orexin A/B has been identified in the dorsal hypothalamic area and LHA. However, the effect of nesfatin-1 signaling with these receptors on ghrelin, leptin, asprosin and orexin A levels is unknown. For this reason, it is important to determine the effect of nesfatin-1's interaction with CRH1, CRH2, oxytocin and kisspeptin receptor antagonists on ghrelin, leptin, asprosin and orexin A level. There are six groups in total: sham (control) group, nesfatin-1 group, nesfatin-1 + p234 group, nesfatin-1 + antalarmin HCl group, nesfatin-1 + astressin2B group, nesfatin-1 + atosiban group. Each group has 5 animals. After the experimental application, the animals were sacrificed and serum ghrelin, leptin, asprosin and orexin A levels were measured by ELISA. Serum asprosin levels did not differ statistically. Serum orexin A levels were found to be low in nesfatin-1+p234, nesfatin-1+antalarmin HCl, nesfatin-1+astressin 2B (p<0.001) and nesfatin-1+atosiban (p<0.01) groups. Nesfatin 1+p234 group had low serum ghrelin level (p<0.05). Nesfatin-1+ p234 group had high serum leptin level (p<0.01). In conclusion, in our study, nesfatin-1 decreased serum ghrelin and orexin A levels and increased serum leptin levels. It did not affect the serum asprosin level. These results showed that nesfatin-1 can affect the peptides involved in the fasting and satiety process.
