Investigation of MDM2 and RYBP gene expression levels in colorectal cancer

Abstract (EN)

Colorectal cancer (CRC) is known as a major cause of morbidity and mortality throughout the world including Turkey. Murine double minute 2 (MDM2) an oncogene is a critical negative regulator of the tumor suppressor p53. Ring1 and YY1 binding protein (RYBP) is a member of the Polycomb group (PcG) and regulates cell growth through PcG-dependent and -independent mechanisms. The objectives of this study were to investigate the expression of MDM2 and RYBP genes in normal and tumor colon / rectum tissues of the Turkish patients diagnosed with CRC. Also, to test the relation of MDM2 and RYBP expression with clinico pathological features such age, gender and to elucidate the correlation between MDM2 and RYBP expression in CRC tissues. Paired tissues tumor-normal (T-N) from 43 patients with CRC included in this study were examined by using qRT-PCR technique. Statistically there was no significant difference between normal tissues (3.60 ± 2.56) and tumor tissue (3.84 ± 2.70) in the terms of RYBP gene expression (p> 0.05). The same result for MDM2 expression was found in which no significant difference between normal tissues (3.20 ±1.79) and tumor tissues (3.36±2.16), (p> 0.05). None of the pathological features measured with the alteration (high and low) of gene expression for both RYBP and MDM2 were had a relation. Also, there was no correlation between the RYBP and MDM2 gene expression (r = 0.158, p = 0.308). Taken together, these data suggest that despite the fact that MDM2 plays as an oncogene and RYBP plays as tumor suppressor gene or maybe as oncogene depending on the tumor type, but both genes have no role in our patient's outcome.
