Abstract (EN)

Antioxidant Effect of Curcumin Against Liver and Kidney Tissue Oxidation of Irinotecan Introduction: Irinotecan is a semisynthetic, water-soluble derivative of the alkaloid called camptothecin, a topoisomerase I group antineoplastic, immunosuppressive drug and is converted by carboxyesterase into its active metabolite SN-38. Curcumin is a polyphenol antioxidant compound produced from castor saffron root. This study was planned to show the liver and kidney side effects of irinotecan and to show the protective effects of curcumin against these effects. Material and Method: 40 Sprauge-Dawley rats were used in the study. (n = 10). Groups; Control, IR, IR + CUR, CUR. TBARS and antioxidant enzymes levels (SOD, CAT, GSH, GPx) were measured in order to evaluate the antioxidant level of the cell as a biochemical evaluation. Hematoxylin-Eosin (H-E) staining method was used for histological light microscopic evaluation. Results: It was observed that liver and kidney tissue samples of rats in the control and Cur groups were in normal histological appearance in light microscopic examination. Congestion, eosinophilic, vascular congestion, hemorrhage, vacuolization, edema and sinusoidal dilatation were observed in the liver tissue in the IR group. In the IR + CUR group, it was observed that histopathological damage to the liver tissue was significantly reduced. Vascular congestion, edema, hemorrhage and glomerular degeneration were observed in the renal tissue of the IR group. In the IR + Curcumin group, it was observed that histopathological damage to the kidney tissue was significantly reduced. Biochemically, it was determined that TBARS level increased significantly in IR group compared to all other groups, and curcumin supplementation significantly decreased TBARS level. It was observed that IR treatment significantly decreased GSH, SOD, GPx and CAT levels, and curcumin supplementation statistically increased all of these parameters. Conclusion: It has been observed that irinotecan has toxic effects on the liver and kidneys, and curcumin significantly reduces these toxic effects.
