Investigation of neuroendocrin effects of irisin hormone on pubertal transition and reproductive system by in vitro methods

Abstract (EN)

It is known that reproductive health, which is necessary for the reproduction of living organisms and the maintenance of their generations, is nowadays impaired by the effects of many genetic or environmental factors. Puberty is a developmental process in which sexual maturation is completed and sufficient reproductive capacity is reached. This process starts with the activation of the gonads by a complex neuroendocrine mechanism including various hormones and neuromodulators which are stimulatory and inhibitory at the central level. Especially kisspeptin, GnRH and GnIH hormones play an important role in this process at the central level. The aim of this thesis is to investigate the possible effects of irisin hormone, which is known as an exercise hormone, on kisspeptin, GnRH and GnIH secretions which have important roles in pubertal period. For this purpose, the effects on hormone secretion and intracellular calcium signalling were examined in immortal cell lines secreting kisspeptin, GnRH and GnIH by in vitro methods. In addition, with the increase in the number of diseases accompanied by neuronal damage in recent years, the neuroprotective activity of irisin hormone was also investigated by performing viability test in these cell lines. Within the scope of the study, it was firstly determined that irisin increased GnRH and kisspeptin secretions in immortal cell lines in a dose- and time-dependent manner after irisin administration, while GnIH secretion decreased statistically significantly (p<0.005). In the study on intracellular calcium signalling, no effect of irisin hormone was observed. Finally, it was determined that irisin hormone had a statistically significant neuroprotective effect in cell viability test studies (p<0,005). All these findings indicate that irisin has an effect on the neuroendocrine reproductive axis and may have an important role in the treatment of neuronal damage disorders as well as sexual dysfunctions.
