Özet (EN)

Everyday, the number of immunocompromised patients increases because of using wide spectrum antibiotics, making more organ transplantations; also in these patients, candidiasis are increased. The immunological changes which occur during the Candida infections have been effected lots of factors. Because of this, we aimed to find the changes of cytokine profile which occur during candidiasis. We used ELISA method which determines the cytokine profiles such as TNF- , IFN-, IL-2, IL-4, IL-1, IL-10 in mononucller cells from the spleen of infected mouse. C. albicans was given the mouse, thus infection model of candidiasis was completed. Also, control group was formed for comparision with infected group. The spleen mononucleer cells which were taken from the infected mouse cultured in vitro and assessed the cytokine profiles. Also, the cells stimulated with LPS (lipopolysaccaride). The mononucleer cells which were taken from the infected mouse expressed the proinflamatuar cytokines such as IFN-, TNF- and IL-1 which levels were increased. Another proinflamatuar cytokine IL-2 levels were observed an increased for 24 hour and decreasing for 48-72 hour. Antiinflamatuar cytokines such as IL-4, IL-6 and IL-10 levels were observed decreased, because they are expressed during the acute infection period. Accordingly, we think that cytokines are important to relation of immune cells during the candidiasis and determining the profiles of cytokines accommodates to protect from Candida infections and improved new alternatives in treatment.
