Demonstrating the expression of IFN-γ, TNF-α, IL-10 with revers transcriptase polymerase chain reaction in rat candidiasis model

Özet (EN)

Everyday, the number of immunocompromised patients increases because of using wide spectrum antibiotics, making more organ transplantations; also in these patients, candidiasis are increased. The immunological changes which occur during the Candida infections have been effected lots of factors. Because of this, we aimed to find the changes of cytokine profile which occur during candidiasis. We used RT-PCR method which determines the cytokine profiles such as TNF-, IFN-, IL-10 in kidneys of infected rat. C. albicans was given the rats, thus infection model of candidiasis was completed. Also, control group was formed for comparision with infected group. The kidney tissues which were taken from the infected rats, the culture results were positive. We showed rat?s kidneys? candidiasis with RT-PCR. Also, the existence of infection was confirmed with spesific primers to fungus. In control group, the results were like these: TNF- % 60 (6/10), IFN- % 60 (6/10) and IL-10 % 10 (1/10). In infected group, the results were like these: TNF- % 91,6 (11/12), IFN- % 100 (12/12) and IL-10 % 75 (9/12). Accordingly, we think that cytokines are important to relation of immune cells during the candidiasis and determining the profiles of cytokines accommodates to protect from Candida infections and improved new alternatives in treatment.
