The effects of platelet rich plasma (PRP) and sevofluran anesthesia in rats with ichemia reperfusion in the liver

Abstract (EN)

The main purpose of this study is to investigate the effects of PRP application and sevoflurane anesthesia on liver ischemia-reperfusion injury with histopathological and biochemical parameters. In the study, 36 Long Evans rats were used and divided into 6 groups. The first group (group D) is the donor group, and blood was taken from the abdominal aorta and sacrificed in order to create PRP. Abdominal laparotomy was performed in the control group (group K), liver tissue was removed and the rats were sacrificed 2 hours after the procedure. In Group IR, Group IR-Sevoflurane, Group-PRP, Group IR-PRP-Sevoflurane groups, the porta hepatis was explored and clamped after abdominal laparotomy; 2 hours of ischemia and 2 hours of reperfusion was applied afterwards. PRP was administered intraperitoneally 30 minutes prior to ischemia to IR-PRP and IR-PRP-Sevoflurane groups and 2.3% sevoflurane anesthesia was administered to IR-S and IR-PRP-S groups during ischemia and reperfusion. Liver tissue samples were examined under the light microscope by staining with Hematoxylin-Eosin (H&E); Malondialdehyde, Catalase, Glutathione-s-transferase and Paraoxonase values were measured in tissue samples for biochemical analysis. In the histopathological examination, hydropic degeneration, sinusoidal dilatation, mononuclear cell infiltration and necrosis in the liver parenchyma were found to be significantly lower in the IR-PRP and IR-PRP-S groups compared to the IR group. There was no significant difference between the groups in the evaluation of the pycnotic core. MDA level and catalase enzyme activity measured in tissue samples were significantly lower in IR-PRP and IR-PRP-S groups compared to IR group; PON enzyme activity was found to be significantly higher in both groups compared to IR group. There was no significant difference between the groups in GST activity. In the IR-S group, there was no significant difference in the findings compared to the IR group. With these findings, it was concluded that PRP application may be effective in preventing experimentally induced IR damage in the liver. More randomized, controlled trials and clinical studies are needed to apply this use of PRP to clinical practice.
