Investigation of cyanobacterial diversity, antimicrobial activity and fatty acid profiles in Karahayit-Denizli spring

Abstract (EN)
In this study, thermophilic cyanobacterial diversity, antimicrobial activities and fatty acid profiles in Denizli Karahayıt red spring water region were investigated. 102 cyanobacteria with different morphologies were isolated from the water samples taken in different months from the Karahayıt thermal water source. The 16S rRNA gene region of the genomic DNA of the isolates was amplified and 19 different band profiles were determined as a result of the ARDRA study. The 16S rRNA gene of representative isolates selected from the ARDRA groups were sequenced with 1492R and 23S primers, and the sequence results were analyzed with the BLAST program. The most closely related sequences were determined as Pseudanabaena sp. NSJ, Gloeocapsa sp. HG2, Cyanobacterium aponinum HG3, Oscillatoria sp. N9DM, Leptolyngbya sp. UIC 10068, Thermophilic cyanobacterium 900, Phormidesmis molle NIES-2126, Oscillatoriales cyanobacterium YACCYB534, Leptolyngbya sp. Lli18, Mastigocladus sp. TB-4, Gloeocapsa sp. PKUAC-GDTS1-13, Geitlerinema sp. CY20, Leptolyngbya sp. CCALA 945 ve Leptolyngbya sp. Pltlpt2. Metagenome study from culture-independent methods revealed that Bacteroidetes, Chloroflexi, Cyanobacteria, Deinococcus-Thermus, Firmicutes, Planctomycetes, Ignavibacteriae, Proteobacteria, Spirochaetes, Verrucomicrobia and Acidobacteria species constitute the microbial diversity in Karahayıt region. In addition, Cyanobacterium aponinum, Coleofasciculus chthonoplastes and Phormidium etoshii species represented the Cyanobacteria group.Agar block diffusion and microtitration method were used for antimicrobial activity study. Some isolates have showed inhibitory effect against M. luteus, A. brassicola, C. glabrata, C. krusei, C. parapsilosis, C. albicans, A. parasiticus, F. moniliforme, F. solani, P. chrysogenum, P. citrinum and P. notatum by agar block diffusion method and also some isolates haved showed antifungal effects against Candida albicans, Aspergillus parasiticus and Penicillium citrinum with the microtitration method. Derivatized FAME GC-MS method was used for fatty acid study. Fatty acids detected in isolates are myristic acid, pentadecanoic acid, stearic acid, linoleic acid, α-linolenic acid, palmitic acid, palimitoleic acid, 7-hexadecenoic acid, oleic acid and 11-octadecenoic acid. In addition, the percentage of total saturated fatty acids and unsaturated fatty acids was determined.
