Abstract (EN)

This study has been carried out in C.Ü. Faculty of Agriculture, Department of Horticulture and C.U. Biotechnology Research and Application Centre. This study has been conducted analyse the effects of cold pretreatment, TDZ, 2,4-D application on watermelon via anther and ovule culture has been established two different testing.. At first study, cold pretreatment has been applied to flower buds for 0, 2, 4, 6 days at +4 oC. After cold pretreatment, anther and ovule were that isolated and has been cultured in MS basal media which contained 0, 1, 3, 5 mg/l 2,4-D, 0, 0.01, 0.02, 0.04 mg/l TDZ and combination of different doses 2,4-D and TDZ. At second study, 3 mg/l 2,4-D for anther culture and 5 mg/l 2,4-D for ovule culture has been stable in all media and has been searched the effect of 0, 0.01, 0.02 and 0.04 mg/l TDZ. Only callus from anther culture has been obtained at the end of both two studies. In case of ovule culture, it has been observed that ovules get bigger and dark green. This structure has been named as plantlets formations. Then, plantlets formations has been transfered to MS basal media without plant growth regulator. After 3-4 weeks of cultured, plantlets formations became brown and lost their vitality.
