The comparative karyotype analyses and chromosomal nor phenotypes of the Cyprinus carpio (L., 1758) and Leuciscus cephalus (L., 1758) live in Kastamonu Beyler and Germeçtepe lakes

In this study, structure and number of chromosomes were investigated, and karyotypes were determined in Cyprinus carpio and Leuciscus cephalus of the Cyprinidae family that were obtained from the population in Beyler and Germeçtepe lakes of Kastamonu in Turkey. The NOR phenotypes of these species were determined, and the population of each species was compared. A solution of 0.1% colchicine was injected into the abdominal cavity at a ratio of 1-mL per 100-g of body weight. 3.5-4 hours later, the fish were killed and kidneys removed. The hypotonization of the kidney tissue was performed with 0.075 M KCl, fixation was conducted in a mixture of methanol and glacial acetic acid in the ratio 3:1 (v/v). Finally, it was stained in giemsa, which was prepared in a phosphate buffer solution (pH = 6.8). The Cyprinus carpio populations of Beyler and Germeçtepe lakes have a chromosome number 2n = 100. The chromosome formula of the Beyler lake populations was found as NF = 152, 22 m + 30 sm + 48 st-a, in the Germeçtepe population it was NF = 150, 20 m + 30 sm + 50 st-a. For Leuciscus cephalus; in the Beyler population; NF = 80, 18 m + 12 sm + 20 st-a, in the Germeçtepe population, NF = 82, 20 m + 12 sm + 18 st-a. The chromosome number for these two populations was determined as 2n = 50. According to the "One Step" method, heteromorphic differences are shown between the populations of these two fish species.
