Effect of catechin and gallic acid on rat liver steatosis

Abstract (EN)
The effect of different doses of catechins and gallic acid to fatty liver tissue was investigated in fatty liver of male rats formed after high-fat diet administration. For this purpose, 160 +/- g of weight 80 Wistar albino male rats were used. Rats were divided into groups as: Group 1 (K), Group 2 (KY), Group 3 (KY + Kat 10), Group 4 (KY + Kat 50), Group 5 (KY + Kat 100), Group 6 (KY + Gal 10), Group 7 (KY + Gal 50), Group 8 (KY + Gal 100), Group 9 (K-10 weeks) and Group 10 (KY- 10 weeks). In order to determine the steatosis, group 1 and 9 were fed with standard rat chow for 10 weeks. And the other groups were fed with high-fat diet for 10 weeks. After 10 weeks, liver tissues and blood samples of Group 9 and Group 10 were investigated as structurally and biochemically. Group 1 was fed with standard rat chow for additional 6 weeks and Group 2 was fed with high-fat diet for additional 6 weeks. Catechins and gallic acid were given by gavage for 6 weeks to other groups as mentioned above 10, 50 and 100 mg / kg / day. At the end of 16-week experimental period, liver tissues and the blood samples from heart were taken from the rats. The iNOS enzyme activity, TG content, ALT, AST, MDA enzyme activities were increased and SOD and CAT enzyme activities were decreased in Group 10 from 10 weekly groups. GSH levels were not different between the two groups. The iNOS enzyme activity, TG content, ALT, AST, and MDA levels were increased and SOD activity was reduced in Group 2(16w). CAT and GSH activities were not different between groups. iNOS enzyme activities and TG content were decreased in KY + Gal 50 ve KY + Gal 100 groups. AST levels decreased KY + Gal 50 group. MDA levels quite decreased in the KY +Kat 100 and KY + Gal 100 groups. Histologically, 3 degree of steatosis was observed in 10 groups (10w). 3 degree of steatosis was observed. in addition to balloning, granular and vacuolar degeneration , sinusoidal dilation , mononuclear cell infiltration in 2.group(16w). KY + Gal 100 is closest structurally normal group, KY + gal 50 is the regulatory group Ast enzyme activity,KY + .kat 100 and KY + Gal 100 groups were found to have the effect of reducing the lipid peroxidation. We could say, KY + Gal 100 group that reduce liver damage in structural and biochmical level.
