Abstract (EN)

The aim of this thesis study was to develop a new oral drug delivery system with ketoprofen lysine salt loaded with Poly Lactic Acid-co-Glycolic Acid (PLGA) nanoparticles which has analgesic and anti-inflammatory properties and to evaluate in-vitro release studies and characterisation of this system. Ketoprofen is a pain relieving and antiinflammatory agent frequently prescribed in high inflammation conditions such as pain, osteoarthritis and rheumatoid arthritis. Ketoprofen lysine salt is a salt of ketoprofen which has been used for many years because of its antiinflammatory activity. However the solubility of ketoprofen and gastrointestinal side effects are limiting of usage. Lysine salt of ketoprofen solubility is better than normal ketoprofen. Coating with a polymeric nanoparticle as an antiinflammatory and painkiller will reduce the gastrointestinal side effects of the drug and eliminate treatment limitation. This protection, especially by the US Food and Drug Administration (FDA) approved by PLGA as a polymer with a polymer used as polymeric carrier system for many years and will be a very reliable and useful drug. Only two ketoprofen lysine preparations are available in our country. None of them has a controlled release. In the scope of the thesis, it was concluded that ketoprofen lysine-loaded polymeric nanoparticles prepared by solvent evoporation, nanoprecipitation and spray driying methods and based on Lactic-co-Glycolic Acid (PLGA) are effective carriers.
