Pou1f1, prlr, β-lg, gh gene polymorphisms in Ki̇li̇s goats and its relationship with milk yield traits

Abstract (EN)
The aim of this study was to determine POU1F1, PRLR, β-LG and GH gene polymorphisms in Kilis goats and to reveal the relationship between genotypes and lactation milk yield, lactation lenght and average daily milk yields. Totally four elite flocks were obtained for animal material in "The National Breeding Project of Kilis Goats under Farm Conditions" which was supported by General Directory of Agricultural Research and Policy, Republic of Turkey Ministry of Agriculture and Forestry. Accordingly, blood samples were taken from 227 head goats in total and genotypes were determined using PCR-RFLP method. T and C allele frequencies were 67.08% and 32.92%, TT, TC and CC genotype frequencies were 40.83%, 52.50% and 6.67% as a result of restriction endonuclease enzyme AluI for intron 5, exon 6 and 3 'flanking region in POU1F1; C and T allele frequncies were 86.80 % and 13.20%, TT, TC and CC genotype frequncies were 75.42%, 22.35% and 2.23% for RsaI loci 3'UTR region in PRLR; A and B allele frequncies were 59.04% and 40.96%, AA, AB and BB genotype frequncies were 30.34%, 57.86% and 11.80% for SacII loci ekzon 7 and 3'UTR region in β-LG; A and B allele frequncies were 55.08% and 44.92%, AA, AB and BB genotype frequncies were 10.17%, 89.83% and 0.00% for HaeIII loci ekzon 2, intron 2 and ekzon 3 region in GH1; C and D allele frequency were 0.83% and 99.17%, CC, CD and DD genotype frequencies were 0.00%, 1.66% and 98.34% for HaeIII loci ekzon 4 in GH2, respectively. In terms of POU1F1, β-LG and GH1 loci, flocks were not found in Hardy-Weinberg equilibrium (P<0.05), but PRLR loci were found in Hardy-Weinberg equilibrium and the homozigoty icreased in GH2 loci. Differences between genotypes obtained in POU1F1, PRLR and β-LG gene regions on lactation milk yield were significant and milk yields of TC, TT and AB genotypes were higher than other genotypes, respectively (P<0.05, P≤0.05, P≤0.001), but GH1 had no effect on lactation milk yield. When the lactation lenght was examined, it was found that the differences between the genotypes on POU1F1 locus were significant and the lactation lenght of the TC genotype was longer than the other genotypes (P <0.05) and PRLR, β-LG and GH1 loci had no effect on lactation lenght. The difference between the genotypes obtained for POU1F1, PRLR and β-LG genes was statistically significant and the milk yield of TC, TT and AB genotypes were found to be high (P <0.05, P≤0.05, P≤0.001), but GH1 had no effect on average daily milk yield. In this study, it is concluded that POU1F1, PRLR and β-LG genes can be used for Marker Assisted Selection in Kilis goat populations.
