Tissue culture studies in coriander (Coriandrum sativum L.)

This study was conducted in Field Crops and Biotechnology Laboratories of Agricultural Faculty during 2009-2010. The study consisted of two stages. In first stage, transferring coriander (Coriandrum sativum L.) plants into tissue culture and proliferation, in the second stage, growth of embryogenic callus were aimed. In the study, Gamze coriander cultivar was used. Before starting the experiments, the seeds were surface-sterilized and germinated on ½ Murashige and Skoog (MS) medium. In the first stage, using MS as a basal medium, seven different media were used for callus induction. For callus induction, some parameters such as callus weight, callus type and callus color were measured and MS + 1 mg/L NAA + 1 mg/L BA medium was determined as the best medium for callus induction. These calli proliferated on the same callus induction medium were used for second stage of the study in which 29 media were tested for callus size, callus score, callus color and green spot percentage of callus. Although no shoot development was observed on calli, green embryogenic calli were obtained on some media. Almost all calli grown on MS supplemented with 3,0 mg/L BA were greenish in color. As a result, callus growth and subculture in coriander were successfully obtained. In addition, the embryogenic calli which is the first stage of embryogenesis or organogenesis were produced. Testing different growth conditions and new media will be appropriate for an entire shoot development in coriander.
