Molecular characterisation of echinococcus granulosus obtained from isolates of cystic hydatidosis cases

Abstract (EN)
Aim: CE is a parasitic infection that is endemic in Turkey. The identification of different strains of Echinococcus granulosus is important for eradication of the disease. The aim of this study is to determine the different strains that cause the disease in the region because of controlling the infection and to contribute to the control programmes. Material and Method: Between November 2013 and December 2015, CE pre-diagnosed patients who were operated at Inonu University Turgut Ozal Medical Center were included to the study. 53 samples were obtained from the patients and evaluated with direct microscopic examination for protoscolex and hook presence. All microscopically diagnosed specimens were preserved until studying. Genomic DNA was extracted from the specimens and mt-CO1 gene region was amplified by PCR. One-way DNA sequence analysis was performed on 33 specimens with 446 bp band images. Results: Protoscoleces and hooks were observed in all specimens examined by direct microscopy. 446 bp of bands were observed in 33 samples as a result of amplification of mt-CO1 gene region. As a result of one-way DNA sequence analysis of 33 PCR products, all isolates were determined as E. granulosus sensu stricto (G1-G3). Conclusion: Studies for identifying different strains of E. granulosus was reported that the most common type is E. granulosus sensu stricto. In our study, all of the isolates studied were identified as E. granulosus sensu stricto. It would be appropriate to investigate the presence of different strains with more extensive studies. In order to carry out the control studies against CE in Malatya, the biological properties of this type must be well known. Keywords: DNA sequence analysis, Echinococcus granulosus, Malatya, PCR.
