Thesis)Dilek ASILGAZI UNIVERSITYINSTITUTE OF SCIENCEJuly 2006ABSTRACTIn this study, semi-IPN hydrogels containing different compositions ofacrylic acid (AA),Citraconic acid and Chitosan (Cs) were synthesized byfree radical polymerization using ammonium persulphate / sodiummethabisulphite [(NH4)2S2O8 / Na2S2O5] as a radical initiator pair in thepresence of ethyleneglycoldimethacrylate (EGDMA) as a crosslinker.Their swelling behavior was investigated as a function of time, pH andtemperature. The percentage swelling (%S) of all hydrogels wereincreased with time but after 24 hours constant percentage swellingwas observed. The presence of AA in gel matrix influences the swellingpercentages. It is found that P(Cs/AA), which is an example of Semi-IPN?s, shows higher swelling behaviour than the other hydrogels. Thepresence of SA in gel matrix influences the swelling percentages.P(Cs/AA) was determined as the most swollen hydrogel at pH=7,4 and37º C. In the second part of the study, P(Cs/AA/SA)-1, P(Cs/AA) andP(AA/SA) hydrogels which are the semi-IPN?s that gave the best resultswere entrapped to Fluconazole. UV spectrophotometer was used tofollow the release of Fluconazole from the entrapped hydrogels. All ofthe release studies were carried out at pH=4 and 37oC. It has been seenvthat P(Cs/AA/SA)-1 hydrogel released the Flucozanole faster than theother hydrogels, when the release times compared. The release ofentrapped fluconazole kinetic data has found by make using from Ficklaw. Calculated k, n parameters are k=1,40x10-2 , n=0,52 for Cs-PAA;k=1,13x10-3, n=1,16 for Cs-P(AA-co-SA)-1and k=1,26x10-3, n=1,11 forP(AA-co-SA) respectively.Science Code : 201.1.117Key Words : Hydrogel, semi-IPN, acrylic acid ,citraconic acid,chitosanPage Number : 94Adviser : Prof. Dr. Mehlika PULAT
