DOÇ. DR. MELAHATKURTULUŞ ÜLKÜERANAB L M DALI: FARMASÖTIK MIKROBIYOLOJIYILI: 2007STATÜSÜ: YÜKSEK L SANS TEZSHV Extended Spectrum β-lactamases are widely present inthe species of Klebsiella pneumoniae. The mutations that occurs in SHVExtanded Spectrum β-lactamases causes new SHV variants.It?s important that determine the isolates that carrying ofresistance genes using more sensitive molecular techniques besides theroutine microbiological tests. Presence of ESBL in Klebsiella pneumoniae andESBL variants was detected by RSI-PCR technique. Restriction siteinsertion-PCR (RSI-PCR) is a simple, rapid technique for detection of pointmutations at Klebsiella pneumoniae. Mismatch primers were designed todetect mutations affecting amino acids at positions 179 and variants carryingmutation have been showed with only one restriction enzyme.41 of the 53 Klebsiella pneumoniae isolates were digested byHinfI restriction enzyme. 23 of them were determined blaSHV-6, blaSHV-8 andblaSHV-24 variants.
