Investigation Of Colon Polyps And Long Non-coding RNA (lncRNA) Polymorphisms, Senem Yılmaz, Specialization Thesis, Background and Aims: Understanding tumorigenic signaling pathways involving lncRNAs in colon polyps will provide further information on colorectal cancer pathogenesis. Therefore, in our study, we aimed to investigate colon polyps and long non-coding RNA (lncRNA) polymorphisms. Matherials and Methods: A total of 125 individuals, 65 patients with colorectal polyps and 60 controls, were included in the study. Peripheral blood samples of the cases were taken into EDTA tube. SNPs in the blood samples, including CCAT1 (RS67085638), HULC (RS7763881), CCAT2 (RS6983267), CASC8 (RS7014346), PVT1 (RS13281615), PRNCR1 (RS12682421) were analyzed by Fluidigm SNPType method. Statistical evaluation was performed using t test, chi-square test and Mann Whitney U test. Results: As a result of the statistical analysis, no significant difference was found between neoplastic non-neoplastic polyp and neoplastic polyp types in patient and control group comparison of CCAT1 (RS67085638), CCAT2 (RS6983267), CASC8 (RS7014346), PVT1 (RS13281615) polymorphisms (p≥0.05). A significant difference was found between neoplastic and non-neoplastic polyps in HULC (RS7763881) polymorphism (p = 0.014). A significant difference was found between neoplastic polyp types in PRNCR1 (RS12682421) polymorphism (p = 0.026). Conclusions: HULC (RS7763881) polymorphism suggests that AC genotype may be effective in neoplastic polyp transition, and PRNCR (RS12682421) polymorphism may play an important role in transition to carcinoma in dysplasia. However, more studies are needed on this subject in order to understand the underlying mechanisms and to advance in diagnosis and treatment.
