Investigation of miR-564 ve miR-718 expression levels in tissues of patients with colorectal cancer

Abstract (EN)
Colorectal cancer is responsible for approximately one in every 10 cancer cases and deaths. In addition to environmental effects, colorectal cancer is caused by multiple genetic and epigenetic changes. MicroRNAs are endogenous RNA molecules that post-transcriptionally regulate protein-coding genes. Numerous studies have been conducted showing that dysregulated miRNA expression profiles are associated with many types of cancer. The aim of this study was to determine the relationship between colorectal cancer and expression levels of miR-564 and miR-718. In our study, healthy tissue samples with tumor and adjacent to the tumor, which were obtained voluntarily by signing the consent form of 80 patients with colorectal cancer, were analyzed. This study was approved by the decision of the Medical Ethics Committee of Gaziantep University Faculty of Medicine, numbered 2021/83. miRNAs were isolated from tumor and surrounding healthy tissues of colorectal cancer patients, cDNA synthesis was performed from these miRNAs, and qRT-PCR method was used to determine the expression levels of miR-564 and miR-718. As a result of the study, it was determined that miR-564 and miR-718 expressions in tumor tissues were significantly decreased compared to normal tissues (p˂0.05). Consequently, it can be concluded that both miR-564 and miR-718 play a tumor suppressor role in the formation of colorectal carcinogenesis.
