Abstract (EN)

Within the scope of this thesis, the glucose-6-phosphate dehydrogenase enzyme from the compost worm was purified. Biochemical characterization studies of the enzyme such as optimum pH, optimum ionic intensity, optimum activity display temperature, content of KM and Vmax, stable pH, product inhibition were determined. Also, the effects of pesticides such as oxamyl, carbaryl, dinaconazole, metalaxyl, methomyl, carbofuran, simazine, tebuconazole, atrazine, propoxur, 1- naphthol, 2,4-D and metal ions such as Pb(NO3)2, Cr(NO3)2, HgC12, FeS04, Ni(NO3)2, Cd(NO3)2, Co(NO3)2, AI(NO3)3 on the activity of Eisenia Fetida glucose-6-phosphate dehydrogenase activity were investigated. Oxamyl, carboryl, dinaconazole, metalaxyl, metomil show inhibition with IC50 vaules between 7,6-77,12 µM. Pb(NO3)2, Cr(NO3)2, HgCl2, FeSO4, Ni(NO3)2 in inhibition with IC50 in 56-120 µM. In conducted study, Ki constants for substances exhibited inhibition effect was calculated by using the Cheng-Prusoff equation.
