The evaluation of the early term healing effects of resveratrol on corneal wounds in rats

Corneal wounds are frequently seen in clinical surveys of cats and dogs. Mainly their prognose are worse or susceptible by because of the outcome of the theraphies. Oftenly, veterinary medicine benefits from the human medicine for the treatment of this disease with modified methods. In this study, a polyphenol substance phytoalexin known as resveratrol was used to adjunct medical and surgical treatments, presumably reducing the side effects of antimicrobials and benefiting from the antitumour, antioxidant effects of resveratrol substance to improve the recovery and reduce or eliminate the cicatrization which marks an era in both medical grounds within this new treatment method. For this purpose, 6 weeks old 20 male Wistar Albino Rat were used in this study. Rats were divided randomly to two groups which Group 1 was control group, and Group 2 was resveratrol group. On each rat for both groups, right eyes' cornea were ensized centrally for 2-3 mm long using lup magnification (2,5X) and wounds were sutured using10/0 nylon material in simple fashion. Tarsoraphy were made and tobramycin was used for treatment. In experiment group, (group 2) rats were fed additionally with resveratrol (30mg/kg) through gavage feeding and in group 1 rats were fed with food and water ad-libidum only for 11 days. Cases were examined for ophatlmoscopic findings. Following the sacrifice of cases at the 11th day speciments examined histologically. The histopatological examination results showed that 1 cases corneal specimen was unavailable by because of inconvenient slicing during speciment preparation. In each group, 9 right eye of rats' corneal speciment was studied. Blood samples were collected prior to sacrifice of animals and biochemical tests were applied. MMP-9 (Matrix metalloproteinase-9), NO (Nitric oxide), MDA (Malondialdehite), AOS (Antioxidan Status) were measured. According to CTP (Connective tissue prolifetaion) scoring 3: respectively group values were: %11.1 and %0.0 (p<0.05). According to Infiltration of Inflamatuaric Cell (IC) scoring 3: respectively %11.1 and %0.0 (p<0.05). MDA results were respectively: Day 0th and day 11th K and RES groups were measured as 1,61±0,316, 2,854±0,57 2,362±0,513. The increase in Group K was statistically significant (p<0.001). MMP-9 was measured respectively; 1,115±0,197, 2,842±0,368 and 2,437±0,323. Group K increase was also found statistically significant (p<0.01). According to AOS the inter-group difference was statistically significant (p<0.05). RES group's 11th day measurements were so similar to day 0 measurements. As a result, according to biochemical and histopathological findings, resveratrol inhibited the vascularisation in corneal wound healing and retained the transparency of corneal tissue. Resvetratrol showed considerable role by maintaining the healing therefore effected the outcome of the treatment. However, it is seen that extended studies and longer follow-ups are needed better evaluation of the outcomes of resveratrol on the healing of corneal wounds.
