The relationship between TGFß1 gene polymorphism and TGFß1, ADAMTS-4 serum levels in coronary artery disease

Abstract (EN)

Coronary heart disease is a multifactorial disease which is responsible for morbidity and mortality in all of the world and is characterized obstructive aterosclerosis in vessel wall. Although the risk factors that cause atherosclerosis are determined, the pathogenesis of this disease can't be explained completely. Recent years there are supporting datas that low density lipoprotein's excessive deposition in vessel wall is the basic mechanism of atherosclerosis(1-2). In this deposition increasing LDL binding capability of proteoglycans that is the basic structure of subendotelial extracellular matrix is charged. Although the predisposition factors that increase LDL binding to proteoglycans have not been determined yet it is speculated these factors that are manufacturing or destruction of extracellular matrix can be responsible in this process. In this study, it was aimed to investigate the role of TGFß1 and ADAMTS-4 in atherosclerotic patient, which are responsible for structuring of extracellular matrix and associated with atherosclerosis. In this study designed above mentioned aims, from cases referred to cardiology department who have coronary heart disease prediagnosis, 84 case inserted in patient group which have atheroma plaque in coronary angiography and 72 cases which coronary angiograhy is normal inserted in control group. Blood samples from cases were taken for analysis of TGFß1, ADAMTS-4 serum levels and TGFß1 rs1800469, rs1800470 and rs4803455 polymorphisms. According to our results, from TGFß1 triple genotypes CCA genotype is more frequent in the patient. There was no difference between patient's and control's level of serum TGFß1. In patients ADAMTS-4 serum levels were higher than controls and ADAMTS-4 levels increased with lesion percentage and vessel number. ADAMTS-4 levels were higher in diabetic cases as well as TGFß1 correlated with ADAMTS-4 levels in patient. In conclusion, it was demonstrated that ADAMTS-4 may have a major role of physiopathology of atherosclerosis, but the correlation between TGFß1 levels in patients should be supported with expression study. The genotype which is detected more frequent in patients, should be investigated in more patients for determining the development of atherosclerosis
