Pathological and immunohistochemical investigations in encephalitic listeriosis in sheep

Abstract (EN)

In this thesis study, it was aimed to evaluate the lesions formed in Listeria monocytogenes (Lm) infection with neurological symptoms in sheep by pathological and immunohistochemical methods. For this purpose, 10 central nervous system tissues from the archive, which had previously exhibited findings related to listeria infection in terms of histopathological examination, were determined to be positive for the agent Listeria monocytogenes by immunohistochemical staining method. In addition, 10 central nervous system tissues from sheep that did not exhibit any neurological symptoms were used as controls. Immunohistochemistry method was applied to determine the expressions of caspase-3, glial fibrillary acidic protein (GFAP), inducible nitric oxide synthase (iNOS) and 8-hydroxy-2′-deoxyguanosine (8-OHdG) in the brain, cerebellum, pons, medulla oblongata and thalamus regions that were immunopositive for the agent and compared with controls. All preparations were evaluated under light microscope and photographed. As a result of these evaluations, it was thought that caspase-3, iNOS, 8-OHdG and GFAP, which play a role in the pathogenesis of encephalitic listeriosis in sheep, could contribute significantly to the explanation of the pathological mechanisms of the disease and to the scientific basis of the treatment options of the disease, especially 8-OHdG.
