Comparison of the cholorimetric method in the measurementof plasma lactate and sheep test device in sheep

Lactate level is a parameter directly related to the severity of the disease and mortality in systemic inflammatory conditions. The aim of this study is to compare the colorimetric method which is fast and easy to use and the standard method of lactate concentration in sheep. For this purpose, lactate concentration of 54 Chios (30 males and 24 females) were compared with the colorimetric tester and ELISA which was evaluated as a reference method. Lactate concentrations measured by both devices were compared using Shapiro-Wilk normality test, Pearson correlation test, Paired t test, Passing-Bablok regression and Bland-Altman analyzes. As a result of descriptive statistics, average values of 5.47 mmol / L in Eurolyser solo device and 3.52 mmol / L in ELISA device were obtained. Pearson correlation analysis was used to evaluate the two methods. There was a high correlation (0.624). Paired t test was used to determine the difference between the two methods. Significance was determined by Paired t test and t value was found to be -8.29. In Passing-Bablok regression analysis, the equation was formed as Eurolyser mmol / L = -4.75 + 2.58 Colorimetric mmol / L. As a result of the Passing-Bablok analysis, it was concluded that there is a systematic and proportional bias since 95% confidence interval does not include 0 (zero) and 1 (one) values. According to the Blant-Altman analysis, it was concluded that the difference between the two methods was good because the difference in mean lactate concentration was 1.04 mmol / l and 96% (52/54) of the values were within the limits of agreement. In conclusion, based on the data of this study, it can be said that Eurolyser Solo can be used for the evaluation of lactate concentration in Sheep.
