Effects of L-thyroxine on in vitro model of osteoarthritis

Osteoarthritis is one of the well-known causes of disability, characterized by loss of articular cartilage. Considering the pathophysiology of OA, it is seen that the regenerative capacity of damaged cartilage is limited. Therefore, the intervention to be made is of critical importance. Thyroid hormones, one of the main hormones that play a major role in the metabolic activity of cells in any tissue, can be a major focus in regenerative medicine. Thyroid hormones have emerged in many studies as a key regulator of bone, muscle and articular cartilage. To prevent terminal differentiation of chondrocytes by sequential cartilage degeneration, it depends on the precise regulation of the local T3 availability of the cartilage cell by the intracellular deiodinase system. In our study, based on all these regenerative effects of L-Thyroxine, the chondrogenesis ability of L-Thyroxine hormone was questioned in an osteoarthritic environment simulated as an osteoarthritic human chondrocyte culture. Waste tissue from patients who had Total Knee Replacement surgery due to osteoarthritis in the first stage was used as a result of this surgery. Primary cell culture was obtained from waste cartilage. In the main control group, it was left normal without adding anything to the culture. In the negative control group, 10% of Hanks' balanced salt solution was added to the mixture, where no response would be expected. In the experimental group, L-Thyroxine, was added to the cells in 4 different groups as 12.5ng/ml, 25 ng/mL, 50ng/mL, and 100ng/ml. As a result; Chondrocyte counts in each group studied with 3 different plates The mean scores of the total subdivision were 9.33 ± 1.15 for the control group, 12.66 ± 1.52 for the HBBS group, and 5 ± 2.25 ng for the 12.5 ng/ml group, respectively. It was found to be 4.33 ± 1.52 for the /ml group, 6.66 ± 0.57 for the 50 ng/ml group and 8.66 ± 1.15 for the 100 ng/ml group. Significant results were seen between the control group and the HBBS group (p:0.039), the 12.5 ng/ml group (p: 0.031), the 25 ng/ml group (p:0.011) and the 50 ng/ml group (p: 0.039).
