Quantification, method validation and chemometric analysis of some secondary metabolites specific to the Labiatae family by GC-MS

Aim: Turkey has a rich flora in terms of plant species diversity due to its different climate structure and its location at the intersection of three different gene centers. In this wealth, medicinal and commercial plants have a special place. An important family in which the species used for this purpose are collected is the Labiatae family. As a genus, mainly Salvia, Thymus, Stachys, Mentha, Satureja and Origanum come to the fore. In this thesis study, 32 species of Labiatae family were collected and ethanol extracts were prepared, especially from the eastern region of our country. A GC-MS quantification method including some secondary metabolites (flavonoid, diterpene, triterpene and steroid) obtained from various species belonging to the Labiatae family or obtained from the market or in previous studies was developed and the method was validated. Finally, the quantitative analysis results of the species by GC-MS were evaluated chemometrically. Material and Method: Sclareolide, sclareol, ferruginol, cryptanol, 6,7-dehydroroyleanone, suginal, 12-hydroxy abieta-1,3,5(10), 8, 11, 13-hexaene, sugiol, inuroyleanone, 12-demethylmulticaulin, 7α-hydroxy- β-sitosterol, salvigenin, stigmasterol, sitosterol, sinensetin, lupenone, α-amyrin, lupeol, 3-acetyl lupeol, 1a,21a-hydroxy-2-3-(1'1'-dimethyl-dioximethylene) urs-9(11 ),12-diene, uvaol, betulin, pyxinol, lup-(20)29-ene 2alpha-hydroxy-3beta acetate, betulin 3,28-diacetate and 21α-hydroxy,2α,3β-diacetoxy urs-9(11), A quantitative method involving 12-diene compounds was developed and validated by GC-MS. All analytical parameters of this method were determined. In addition, content analyzes of the ethanol extracts of 32 species commonly used among the public were carried out with this developed GC-MS method and the results were evaluated chemometrically. Results: In general, it was determined that the aerial parts of the studied species were triterpene, steroid and flavonoid-rich root extracts were rich in diterpene. In particular, some of the studied species were found to be source species for için-sitosterol, lupenone, salvigenin, stigmasterol, ferruginol, cryptanol, sclareol and betulin compounds.Conclusion: Extracts of the samples of 32 species grown in our country were prepared and content analyzes were made. The species that can be a source for these compounds have been determined, a new method has been validated with GC-MS and the results have been brought to the scientific world.
