Determination of cyanobacteria diversity in Ladik lake by fatty acid analysis and molecular methods

Abstract (EN)

In this study, cyanobacteria diversity was determined by sampling from Lâdik Lake located in the borders of Ladik district of Samsun province. The 16S rRNA gene region of the isolated isolates was amplified with specific primers and the amplification products were cut with MboI restriction enzyme and ARDRA treatment was applied and as a result of the sequence analysis, isolates of Synechocystis sp., Phormidium autumnale, Phormidesmis sp. genus were obtained. Obtained isolates with different ARDRA profiles were extracted using hexane. FAME analysis was applied to the extracts. The antimicrobial activities of the extracts were determined by the MIC (minimum inhibitory concentration) method. In addition, the antioxidant activities of the extracts were determined by looking at the DPPH (2,2-diphenyl-1-picrylhydrazil) radical scavenging activities and the Copper (II) ion reduction based antioxidant capacity (CUPRAC: Cupric Ion Reducing Antioxidant Capacity) results. Antimicrobial activities of Gram-positive (+) bacteria Bacillus cereus ATCC 7064, Micrococcus luteus NRLL 1018, Bacillus subtilis ATCC 6633, Staphylococcus aureus ATCC 25923, Gram-negative (-) bacteria Salmonella typhimurium LT2, Klebsiella luteus NRLL 1018, Espresso ATCC 7003, Espresso bacteria ATCC 27853, yeasts Candida albicans ATCC 14053 and Candida krusei ATTC 34135 were used. Bacillus subtilis ATCC 663, Bacillus cereus ATCC 7064 and Synechocystis sp. L4-11 coded isolate belonging to the genus showed. In the free radical scavenging ability using DPPH, Syenechocystis sp. L3-23 coded isolate and extract of L3-27 coded isolate belonging to the genus Phormidesmis sp. The extract of the L5-2 coded isolate of the genus L5-2 showed the maximum antioxidant potential. The CUPRAC value was determined as 8.9 µg TE/gr of the L3-27 coded extract and 2.5 µg TE/gr of the L5-2 coded extract.
