Abstract (EN)
With the discovery of the chemical contents of medicinal and aromatic plants, it was predicted that these plants could be used as raw materials for medicine, and studies on the biological activities of plants increased. It is known that plants belonging to the Lamiaceae family, which are very rich in polyphenolic compounds, have high biological potential. In the literature, there is not much data on the biological potential of Lamium orientalis Fisch & Mey. In this thesis study, anticancer, antibacterial, and anti-Quorum Sensing (anti-QS) activities of methanol (MeOH) extracts of the root, stem, leaf, and flower parts of the L. orientalis were evaluated. The anticancer activity of the extracts was investigated by using the MTT test on MCF-7 and MDA-MB-231 cell lines. As a result, it was determined that the MeOH extract of the root part with the best anticancer activity was on the MCF-7 cell line, and the IC50 value was calculated as 425,7 µg/mL at the end of 48 hours. The antibacterial activities of the extracts on standard bacterial strains were determined by the broth microdilution method. The minimum inhibitory concentration (MIC) values of the extracts could not be determined against the strains at the concentrations studied. To determine the anti-QS activities of the extracts, violacein pigment production, and biofilm formation on Chromobacterium violaceum ATCC 12472 and Pseudomonas aeruginosa PAO1 strains, respectively, were investigated. However, it was determined that the extracts did not have anti-QS activity at the concentrations studied. This thesis study is a preliminary information about some biological activities of MeOH extracts of the root, stem, leaf, and flower parts of the L. orientalis. It was observed that the MeOH extract of the root part of the plant has anticancer activity potential on the MCF-7 cell line. In addition, it is foreseen that the L. orientalis will contribute to the development of new chemotherapeutic agents.
