Simultaneous determination of levamisole HCL and triclabendazole with HİGH performance liquid chromatography

Abstract (EN)
In this study, it was aimed to develop simultane determination method for veterinary medicine in the form of oral suspension containing Levamisole HCl and Triclabendazole active drug components. In the literature review studies, it was found that there was no analysis by High Performance Liquid Chromatography (HPLC) method for the simultane determination of these two drug substances. Therefore, it was thought that these two substances, which are offered for treatment in combination, are required to be analyzed by HPLC. In the developed HPLC method, pH 8.0 potassium dihydrogen phosphate - acetonitrile 52:48 (v / v) mixture was used as the mobile phase, and Phenomenex Gemini C18 (150x4.6 mm 5 µm) column was used as the column. The substances were monitored with a UV-Vis detector at a wavelength of 247 nm. The two active ingredients were determined simultanously in a 15-minute analysis period with a flow rate of 1.5 mL / min. The developed method was validated and the linearity range was determined as 60-90 µg / mL for Levamisole HCl and 96-144 µg / mL for Triclabendazole. Detection (LOD) and quantitation limits (LOQ) are 1,6 ng / mL and 4,7 ng / mL for Levamisole HCl, and 17 ng / mL and 53 ng / mL for Triclabendazole, respectively. Recovery values were in the range of 98.6-99.2% for Levamisole HCl and 98.7-99.3% for Triclabendazole. RSD values of system precision standard injections are 0.13% and 0.16% for Levamisole HCl and Triclabendazole, respectively. These results were found to meet the acceptance criteria with values of <2.0%. The veterinary medicine in the form of oral suspension were successfully analysed by the developed method.
