The investigation of protective effect of Levosimendan in myocardial ischemia.

In all experiments, the infarct size and hemodynamic parameters (LVDP, LVEDP, ±dP/dt), measured by MP35 recording system, were evaluated and compared among their experiment groups. These groups that were obtained from isolated rat hearts were exposed individually to ischemic postconditioning and reperfusion with levosimendan after a- 30-minute period of ischemia.Firstly, Wistar rats were anaesthesied with thiopental sodium and hearts were put into solution after thorax was immediately opened. Finally, the hearts were mounted on the Langendorff system quickly. Left ventricular pressure was measured with a water-filled ballon. After a 15 minute stabilisation period, the hearts were performed by ischemic postconditioning and reperfused by levosimendan with different groups. After ischemia, the hemodynamic parameters were examined at 30th and the 120th minutes of reperfusion. The heart tissues were dyed with TTC for infarct size measurement.It was also observed that LVDP, LVEDP and ±dP/dt results of reperfusion at 30th minute were smilar to the pre-ischemia period in the groups in which ischemic postconditioning and levosimendan were performed. However at the 120th minute period, the results were reduced in the same groups. No statistically significant diffrence was observed in LVDP and LVEDP results of both 30th and 120th minute period. The infarct size was significantly reduced in levosimendan and ischemic postconditioning groups compared to the control group.The results showed that, levosimendan and ischemic postconditioning may provide cardioprotection over the myocardial ischemia reperfusion injury.Keywords: Levosimendan, Myocardial ischemia, Langendorff
