Abstract (EN)
Carbonic anhydrases (E.C. 4.2.1.1.) are family of zinc metalloenzymes that first purified from mamalian erythrocyte. Carbonic anhydrases catalyze the reversible hydration of carbon dioxide in a two-step reaction to yield bicarbonate and proton. The isoenzymes of human eye are CAII and CA-IV. Carbonic anhydrase inhibitors, which reduce aqueous production with a corresponding decrease in intraocular pressure (IOP), have been used as ocular hypotensive agents for the treatment of glaucoma.In a study of fungies have effect reduce blood sugar and cholestrol, antioxidant, the treating eye diseases, antitumor. We studied the inhibition effect of medicinal mushrooms, beta-D-glucan, lectin, linoleic acid and beta carotene on the carbonic anhydrase.Before, carbonic anhydrase enzymes from human erythrocyte were purifed using affinity gel and all of the investigations were carried out with these enzymes. Later, the obtained beta D-glucan, lectin, linoleic acid and beta carotene on the inhibition effect on human erythrocyte HCA Iand HCA II enzymes were studied. In these studies, it was taken advantage of hydrates and esterase activites for determining of carbonic anhydrase activitates. HCA on esterase activity, lectin, linoleic acid, and beta carotene compounds showed inhibitory effect on the activation effect of beta D- glucan was determined. I50 values were determined by drawing % activity-[I] graphs for drugs showing inhibition effects. For CO2-hydratase activity of compound lectin to Linoleic acid inhibition effect ranged from 2,41 to 2,3 ? M for HCA I and 2,52 to 1,48 ? M with I50 for HCA II. As for esterase activity of, compounds lectin, linoleic acid to beta carotene had inhibition ranged from 4,9, 0,17 to 1,45 ? M for HCA I and 2,7, 1,2 to 0 92 ? M for HCA II with I50.
