The recent discovery of the presence of fetal DNA in maternal plasma and serum hasoffered new approaches to non-invasive prenatal diagnoses. Invasive prenatal diagnosismethods such as Chorionic villus sampling (CVS) and amniocentesis, entail procedure-related risks of fetal morbidity and mortality; however, non-invasive prenatal diagnoseswould not only obviate the procedure-related risks of an invasive prenatal diagnosis, butalso remove the anxiety associated with them on the part of pregnant women intending toundergo the procedure. An example of non-invasive prenatal diagnosis is fetal genderdetermination by isolation of fetal DNA from maternal plasma and serum and analysis withpolimerase chain reaction (PCR). Fetal gender determination to non-invasive prenataldiagnosis is as a ?pre-test? to determine whether invasive prenatal diagnosis should beperformed on a fetus having a risk of an X-linked recessive disorder. If gender of fetus isfemale, the application of invasive methods is unnecessary. By now, along with the improvingtechnology the fetal gender has been determined in different sensitivity by analysis of fetalDNA in maternal plasma and serum.In this study, it has been evaluated the presence of fetal genetic material in theperiferal blood of pregnant women by means of PCR analysis. For this reason, it has beenaimed to show the presence of fetal DNA in total DNA isolated from serum samplingdecomposed the periferal blood of pregnant women with the help of conventional and real-time PCR analysis with Y chromosome specific sequences (DYS14 and SRY). We onlyreached 25 cases? results in 40 cases examined. All statistical analysis made with these 25cases. Our study showed the presence of fetal DNA in maternal serum by means ofconventional PCR analysis of DYS14 and SRY sequences in accuracy %80 and %53.3respectively .Key words: Prenatal diagnosis, PCR, fetal DNA, circulating DNA, maternal serum, fetal sexdetermination.
