Özet (EN)

SUMMARYTo Determine the relationship between Hypoxia - İnduciblefactor (HIF)-1 alpha Gene Polymorphism and Breast cancerBreast cancer is one of the most common type of canceramong women and many factors existent as a reason of this diseases. The called `HIF-1α? gene is considered as an effective gene during theformation of many types of illnesses- heart diseases and apoplexy- whichcauses to a hypoxic environment. Especially hypoxic tumor cases givepossibility to tumors to grow up and convert genes to a more aggressivephenotype and activate. Hence, the activation of HIF-1α genes arecorrelated with many tumors and oncological pathways and it is thoughtthat its polymorphism increase tendency for many types of cancer. Theaim of this study is to investigate the risk correlation of HIF-1α genepolymorphisms and breast cancer.102 persons suffering from breast cancer and 102 healthypersons as control group are included in this study. DNA isolation iscarried out with complete blood DNA extraction method. Genotypes aredetermined with PCR and DNA sequence analyses methods. In conclusion, no evidence of a relationship, that wouldconstitute a risk factor, was found when the breast cancer patients and thecontrol group of the Turkish population were compared in terms ofC1772T, G1790A and C111A polymorphisms of the HIF-1α gene.Furthermore, it has been shown that the CC and CT+TT genotypes incase of C1772T polymorphisms do not alter these results of the clinic orhistopathologic properties for patients. Keywords: HIF- 1 alpha, breast cancer, PCR, polymorphism, risk factor
