Abstract (EN)

In studies in the literature focused on the effects of TREM2 on inflammation. In two works, were examined indirectly TREM2 expression in monocytes relationship with immune response against lung cancer The aim of this study, investigation of the effects of TREM2 on proliferation, cell death / cell cycle on breast cancer cells and clarirfy its role in cancer development. in vitro study was carried out for this purpose. Before this study one work have been made about this topic with glioblastoma cells.In this study, MCF10A immortalized normal breast cells, SKBR3 and MDA -MB-231, MCF7 breast cancer cells were used. Western Blot was used for analysis the expression of TREM2 in the third cells. Cell proliferation was tested by MTS, colony forming ability was tested by colony forming assay in the cell treated with siRNA. Trypan Blue Exclusion assay was used for determination role in the cell death.TREM2 expression was determined in MDA-MB-231, MCF7 and SKBR3 cells. In the cells treated with siRNA, cell proliferation and colony forming ability had reduced. Cell death was occured %60-65 in MCF10A, %45 in MDA-MB-231, %35-40 in SKBR3. A significant difference was not detected in the cell cycle. In this study, for the first time TREM2 expression was analyzed in breast cancer cells and mammary epithelial cells in vitro. TREM2 enhancing suppression was determined that a reducing effect on cell proliferation and cell death. This study indicated the expected effects of the drugs used in cancer treatment. As a result of this study it was determined to be a new hope for the treatment of cancer TREM2 targeting.
