Abstract (EN)

A disorder in autophagy can cause very pathologies, especially cancers. In this study we wanted to search the expression rates of autophagic gene family in cancer cells. Our purpose is to detect cancer before clinic changes with gene analysis to be able to make early diagnosis and to help molecular bases of gene treatments for cancer in future. We collected normal and cancer tissues of 71 patients in operations. From fresh tissues samples, we derivated RNA with "Qiogen" kits in Medical Biyology and Genetic laboratories. With Real Time Polymerase Chain Reaction (PCR) which have high capacity of nano tecnology, research of gene expression was done with BioMark™ HD System. Expression levels of ARSA, CXCR4, HGS, HPRT1, HDAC1, HTT, SNCA, SQSTM1 and INS autophagic genes were measured. We analysed Copy counts, Ct values and peaks off gene expressions individually, with 2–ΔCt method and statistical analysis were done. 71 female patients were included. The main age was 47,6. Pathologic diagnosis was invasive ductal carcinoma for 62(%87,3) patients. All stages existed in study. There was an increase of expression in tumor tissue for CXCR4, HPRT1, HDAC1 genes and a significantly decrease for ARSA, HGS, HTT, SNCA, SQSTM1, INS genes. It is clear that autophagy has a role in cancer development and progression as well as apoptosıs. We think that more researches about this mechanism can help for early diagnosis and treathment of cancer.
