Investigation of the effect of chlorambucil and tamoxifen on breast and ovarian cell lines

This study aims to examine the cytotoxic, morphological and molecular effects of Tamoxifen and Chlorambucil on MDA-MB-231 and HeLa cell lines and is a preliminary study in the development of cancer drugs in further studies. Cancer is in the second group of diseases that cause death after the most common heart diseases in the world. Various treatment methods such as surgery, chemotherapy and radiotherapy are used in cancer treatment. However, considering the side effects of the treatments, it is important to develop new drug treatments. Tamoxifen is a non-steroidal drug from the group of anti-estrogens. In recent years, Tamoxifen has been used for both breast cancer treatment and chemoprevention. Chlorambucil drug is used in Chronic Lymphocytic Leukemia (CLL), where the bone marrow produces too many abnormal white blood cells, in Hodgkin's lymphoma, a type of cancer originating from lymphatic system cells, in Waldentsrom's macroglobulinemia in which B-lymphocyte white blood cells proliferate excessively and cause abnormal protein release in the blood. It is used to treat ovarian, breast and testicular cancer. Based on this hypothesis, it was aimed to investigate the effects of Tamoxifen and Chlorambucil drugs on MDA-MB-231 and HeLa cell lines. Optimization studies were carried out at certain dose intervals to determine the toxic effects of drugs on cells. In the experiments conducted in this thesis, it was observed that tamoxifen was effective on cells when applied alone, but its effects on 3D MDA-MB-231 and HeLa cell lines were less than studies in the literature. On the other hand, it has been observed that the specific toxic effect of chlorambucil is stronger than tamoxifen.
