Abstract (EN)

SUMMARY p53 Gene Mutation Analysis in Cases with Bladder Cancer P53 gene is a tumor supressor gene mutated in approximately 50% of human malignancies. It performs its tumor supression effect by regulating many genes and canalizing them for cell cycle regulation, DNA repair and apoptosis by many pathways. p53 has the property of being the most mutated tumor supressor gene in bladder cancer. Due to this, it may be an important biomarker in bladder cancer prognosis. In this study, DNA was isolated from bladder tumor tissue samples of 22 cases diagnosed to be urothelial carcinoma. After then, 5th, 6th and 7th exons of p53 were amplified by PCR method and p53 mutation analysis was performed by enzyme restriction in 5th and 7th exons. In 45% of cases performed mutation analysis, p53 gene mutation was detected. 60% of mutations were in 5th exon, codon 175 and 40% of mutations were in 7th exon, codon 249.
