The effects of PPAR GAMMA 2 PRO12ALA polymorphism on lipid profile and adipocyte fatty acid binding protein level in patients with metabolic syndrome

Abstract (EN)
Metabolic syndrome which appears with the interaction of genetic and environmental factors is an important morbidity cause. Adipocyte fatty acid binding protein (A-FABP) is accepted as a marker of the obesity and metabolic syndrome in many populations and expression of A-FABP regulates by the PPAR?. The aim of this study was to examine the effects of PPAR?2 Pro12Ala polymorphism on A-FABP and lipid profile.One hundred ninety six patients diagnosed metabolic syndrome and 98 healty controls were included in this study. Serum glucose, total cholesterol, HDL-C, LDL-C,VLDL-C, triglyceride, A-FABP and insülin were measured in MS group and control group. PPAR ?2 Pro12Ala polymorphism was analysed by PCR-RFLP method.When the MS group was compared with the control group, it was determined that glucose, cholesterol, LDL-C, VLDL-C, triglyceride, insülin, HOMA-IR and A-FABP were higher, HDL-C was lower in MS group. There was a positive correlation between A-FABP and body weight in controls and positive correlations between A-FABP and BMI, waist circumferance were observed in MS group.In control group, 13 Pro12Ala heterozygot, 82 Pro12Pro homozygots and 3 Ala12Ala homozygots were determined and 163 Pro12Pro homozygots and 33 Pro12Ala heterozygots were determined in MS group. No significant differance was seen in glucose, cholesterol, LDL-C, HDL-C,VLDL-C, triglyceride, insülin, AFABP and HOMA-IR between the three genotypic groups in control group and between two genotypic groups in MS (P>0,05).When examined BMI and waist circumferance, there was not a significant differance between the genotypic groups in control group but BMI and waist circumferance were higher in Pro12Ala heterozygot individuals than Pro12Pro homozygot individuals in MS (P<0,05).In conclusion A-FABP is increasing in MS but there were not effects of PPAR?2 Pro12Ala polymorphism on A-FABP levels and lipid profile. PPAR?2 Pro12Ala polymorphism is associated with increased BMI and waist circumferance in MS.Key words: Metabolic syndrome, A-FABP, PPAR?, Pro12Ala, polymorphism
