TEZ KONUSU: RAPID IDENTIFICATION WITH POLYMERASE CHAINREACTION OF METHICILLIN RESISTANT STAPHYLOCOCCUSAUREUSÖĞRENC N N ADI ve SOYADI: MELTEM ÇAYTEZ DANIŞMANININ ADI VE SOYADI: YRD. DOÇ. DR. SUMMARYS. aureus is one of the most encountered agent in which theinfections originated in hospitals and also in society. Nowadays, frequency ofMRSA is increased and it shows diversity, according to countries, districts,hospitals and also departments in the hospitals. There are severalmechanisms in S. aureus that is responsible of methicillin-resistance. It isconvenient using traditional and moleculer methods in determination ofmethicilline-resistance.In this study, disk diffusion and PCR methods are used indetection of methicilline-resistant S. aureus.The samples provided from microbiology laboratory of theMinistry of Health, Dışkapı Education and Research Hospital of ChildrenDiseases, Dışkapı Yıldırım Beyazıt Education and Research Hospital, andtotally 92 samples are studied which are determined by hospital infectionscontrol comittee Faculty of Medicine, Gazi University.The sensitivity of methicilline of the collected clinical samples, isdetected by disc diffusion method. The DNA is provided from S. aureusstrains by boiling method. The gene mecA is amplify by using PCR method.The gene mecA is detected by PCR method in 71 of 92 MRSAstrains which is used in the study.Using PCR method, determination of distribution ofmethicilline-resistance at the other hospitals can be beneficial for preventionof infections.
