The effect of cannabinoids on end organ injury induced by mesenteric ischemia reperfusion

Abstract (EN)
The aim of this study was to investigate the effects of cannabinoids on damage in distant organs after mesenteric ischemia/reperfusion (I/R). The mesenteric I / R model was formed in rats with a 30-minute ischemia and 24-hour reperfusion in the superior mesenteric artery. Monoacylglycerol lipase (MAGL) inhibitor JZL184 (8 mg/kg ip), non-selective cannabinoid receptor agonist cannabidiol (CB; 10 mg/kg, ip) and selective CB2 receptor agonist JWH133 (10 mg/kg, ip). Tissue samples were taken to examine the effects of drugs on apoptosis pathways (caspase 3, 8, 9) and tissue injury. In the histopathological examination, JZL184 significantly decreased alveolar congestion, hemorrhage, neutrophilic infiltration and alveolar wall thickness in lung tissue. JWH133 significantly decreased neutrophilic infiltration and alveolar wall thickness, but decreased alveolar congestion, but this decrease was not statistically significant. In intestinal mucosal damage in intestinal tissue, CB and JZL184 decreased epithelial damage compared to ischemia group but this decrease was not statistically significant. In immunohistochemical examination, CB showed a statistically significant decrease in expression of caspase 3, 8 and 9, and expression of JZL184 caspase 8 in lung tissue. In the intestinal tissue, CB and JZL184 significantly reduced the expression of caspase 3, 8 and 9. JWH133 decreased the expression of caspase 3 and 8 in intestinal tissue but this decrease was not statistically significant. In this study, it has been shown that cannabinoids have protective effect on tissue damage and apoptotic pathways in acute lung and intestinal damage caused by mesenteric I / R. These positive results will enable cannabinoids to be considered a potential drug in lung diseases.
