Molecular analysis of sirt genes and sirt related genes in multiple sclerosis

Abstract (EN)

Multiple Sclerosis (MS) is a chronic disease that is evolved via autoimmune mechanism triggered by environmental factors in genetically sensitive individuals. It continues throughout the human life and could be observed as stiffness in multiple places of central nervous system (CNS), which is characterized with the axon damage and demyelination; that is, inducing the solidified, and sclerosis tissue in brain and/or spinal cord by causing the emergence of infarct. There are many studies on multiple sclerosis, however; the reason of MS has not been certainly known yet. In this study, the expressions of SIRT (sirtuin) gene family prominent because of its relation with the several metabolic events like human metabolism, ageing and cancer and proved to prevent the Alzheimer disease -a neurodegenerative disease like MS-in 2010 and the related 65 genes were examined. In our study, the gene expression differences between 95 MS diagnosed patients and 95 healthy individuals without any neurodegenerative disease was revealed via Fluidigm Dynamic Array. Evaluating the results by statistical calculations, significant changes in the expression levels of 11 genes were observed between the healthy and the patient group. Although its diagnosis was made 140 years ago, the data revealed by this study, while filling a gap in the literature, will contribute to understand the biological structure of MS disease, any treatment method of which has still not been found. SIRT family and several interactions in which it is involved are thought to give hope for the diagnosis and the treatment of MS disease.
