Abstract (EN)

N-nitrosoethylamine are one of the important groups of carcinogens frequently present in human environment and food chain.Ellagic acid, a polyphenol found in berries, grapes and nuts, exhibits important health promoting effects via its antioxidant, antiproliferative, chemopreventive and antiatherogenic activities.The aim of this study is to investigation of oxidan-antioxidan systems in rat kidney tissues treated with N-nitrosodiethylamine and proflactic/protective effects of ellagic acid in these rat kidney tissues.For this reason, the animals were randomly divided into five groups. Each group consisted of six animals. The first group included control group: they were fed with only standard rat diet and tap water for 10 weeks. The second grup included NDEA grup: in this grup rats were treated with 200 mg/kg single dose N-Nitrosodiethylamine (NDEA) as IP at 15. day of the experiment. The third group included ellagic acid group: rats were treated with 25 mg/kg every other day dose EA via gavage, and received was started of the 15. day of the experiment. The forth group included NDEA + Ellagic acid: rats were treated with synchronised intraperitoneal NDEA plus ellagic acid 25mg/kg every other day via gavage at 15. day of the experiment. The five group included EA+NDEA grup: rats were treated with 25 mg/kg/ every other day via gavage ellagic acid in start the first day of the experiment and then NDEA received at the above-mentioned dose at 15. day of the experiment.In this study, there was a significantly increase (P= 0,009) in levels of MDA in kidney tissues at NDEA group as compared to control group, and in the same time there were significantly decrease (P= 0,009) in levels of MDA in kidney tissues at EA group as compared to NDEA grup.It was a significantly decrease (P=0,014) in levels of MDA in kidney tissues at EA+NEDA group as compared to NDEA group.There were significantly increase (P=0,462) in activities of GSH-Px in kidney tissues at EA+NDEA groups as compared to NDEA group.In this study there was a significant increase (P=0,016) in activities of GST in kidney tissues at EA group as compared to NDEA group.In conclusion, we have shown the addition of NDEA to rats increased lipid peroxidation. Ellagic acid decreased the MDA levels in NDEA treated animals. Ellagic acid improved SOD, CAT, GSH-Px ve GST activities in kidney. Hence we suggests that EA has a protective effect aganist kidney toxicity caused by NDEA. This protective effect of EA seems to be closely involved with the suppression of oxidative stress.
