Abstract (EN)

In the study, the biomolecular interaction between DNA-targeted anticancer drug, Dakarbazine (DCB), and DNA was determined by electrochemical method using single-walled carbon nanotubes (SWCNT) modified pencil graphite electrodes (PGE). In this context, the interaction between DCB and DNA with SWCNT modified electrodes was investigated by differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. Voltammetric analysis measured DCB and guanine oxidation signals before and after interaction, respectively. Experimental conditions such as DCB concentration and interaction time of DCB with DNA have been optimized. In addition, the interaction of DCB with nucleic acids was studied using PCR samples and with electrochemical methods and gel electrophoresis technique. In conclusion, the interaction of anticancer drug, DCB, on the surface of SWCNT modified PGE, with nucleic acids was investigated in this study by using electrochemical method, and accordingly the results were compared to the one of electrophoresis method. It has been concluded that the electrochemical assay of DCB-DNA interaction could be performed selectively, sesitively and cost- effectively by using SWCNT- PGE.
